#1 - Acute necrotic hepatotoxicity caused by Lantana camara L. ingestion in dairy cattle

Abstract in English:

This study describes an outbreak of acute necrotic hepatopathy associated with spontaneous poisoning by Lantana camara L. in dairy cattle. A herd of 15 cows and heifers was introduced into a native pasture with limited food supply, and, sixteen days later, eight animals had spontaneous nasal hemorrhage, fever, lethargy, jaundice, and dry, dark stools with mucus and blood. The clinical course varied from two to five days. In the pasture where the cattle were kept, abundant adult specimens of L. camara L. with evident signs of consumption were observed. In total, seven cattle died and necropsy was performed in three of them. All animals had moderate jaundice, hemorrhage in the subcutaneous tissue and on the surface of different organs. The liver was slightly enlarged, with orange discoloration and enhanced lobular pattern. Histologically, multifocal areas of coagulative necrosis of hepatocytes in the centrilobular area, occasionally extending to the midzonal area, were observed, as well as marked hepatocellular degeneration and prominent cholestasis. The current study suggests that L. camara L. poisoning should be considered a differential diagnosis of acute and necrotic hepatotoxicity in cattle, despite the absence of photosensitization.

Esse estudo descreve um surto de hepatopatia necrótica aguda associada a intoxicação espontânea por Lantana camara L. em bovinos leiteiros. Um lote de 15 vacas e novilhas foram introduzidas em um piquete com campo nativo, com escassa oferta de alimento. Após dezesseis dias, oito animais manifestaram epistaxe, febre, apatia, icterícia, fezes ressecadas e escuras com muco e sangue. A evolução do quadro clínico variou de dois a cinco dias. No piquete em que os bovinos estavam alojados havia grande quantidade de L. camara L. com sinais evidentes de consumo. No total, sete bovinos morreram, e destes, o exame post mortem foi realizado em três. Os bovinos exibiam moderada icterícia, hemorragias no tecido subcutâneo e na superfície de diferentes órgãos. O fígado estava discretamente aumentado, com coloração alaranjada e padrão lobular evidente. As lesões histológicas consistiam em acentuada necrose de coagulação de hepatócitos em região centrolobular, por vezes se estendendo a região mediozonal, além de acentuada degeneração dos hepatócitos e evidente Colestase. O presente trabalho alerta para que intoxicação por L. camara L. seja levada em consideração nos diagnósticos diferenciais de hepatotoxicidade necrótica aguda em bovinos, mesmo sem indícios de fotossensibilização.

E-mail: furlan@ufmt.br Epidemiological features, clinical signs and pathological findings of a spontaneous acute poisoning by Pteridium arachnoideum and Pteridium caudatum in cattle in the State of Mato Grosso, Brazil, are described. The plant distribution, magnitude and some aspects about the pasture invasion by P. arachnoideum and P. caudatum in the State are also described. Pteridium spp. were found in 83 farms from nine counties and 22 of them were submitted to taxonomic identification. P. arachnoideum was found in 22 farms and in two farms also P. caudatum. Deforestation and burning appear to be associated with pasture invasion by Pteridium spp. On the farm where the poisoning occurred 306 cattle were introduced into a pasture of Brachiaria brizantha intensely invaded by P. arachnoideum and P. caudatum. Twenty two cattle were poisoned by the plants and died. The main clinical signs were hemorrhages, high fever, apathy, fatigue, weakness and recumbency. Gross findings included variable degrees of hemorrhages in several organs and cavities with occasional infarcts. The most consistent histological finding was marked bone marrow aplasia. Epidemiological, clinical and pathological findings associated with the taxonomic identofication of the plant support the diagnosis of acute poisoning by P. arachnoideum and P. caudatum in cattle. The increasing rate of deforestation and frequent burning in Mato Grosso can contribute to this disease which becomes a major cause of economic losses to livestock production in the State.

Pteridium spp. foi observada em 83 propriedades de nove municípios de Mato Grosso e deste total, amostras de 22 propriedades foram coletadas para identificação taxonômica. Em 22 propriedades identifica-se P. arachnoideum e em duas dessas havia também P. caudatum. O desmatamento e a realização de queimadas parecem estar relacionados com a invasão de Pteridium spp. nas pastagens. Na propriedade em que ocorreu a doença, 306 bovinos foram introduzidos em uma pastagem formada por Brachiaria brizantha intensamente invadida por P. arachnoideum e P. caudatum e desses 22 bovinos adoeceram e morreram. Os principais sinais clínicos foram febre alta, apatia, fadiga, fraqueza e decúbito. Na necropsia havia graus variáveis de hemorragias em vários órgãos e cavidades e infartos ocasionais. Microscopicamente, a alteração mais importante consistiu em aplasia severa da medula óssea vermelha. Os achados epidemiológicos, clínicos e patológicos associados à identoificação taxonômica da planta fundamentam o diagnóstico de intoxicação aguda por P. arachnoideum e P. caudatum em bovinos. O crescente desmatamento e a frequente utilização de queimadas em Mato Grosso podem contribuir para que esta doença venha a ser uma importante fonte de prejuízos para a pecuária do Estado.


#3 - Acute poisoning by trichlorfon in goats given a therapeutic dose, 34(2):114-118

E-mail: wdzlopes@hotmail.com The present study describes an outbreak of trichlorfon poisoning in goats from the State University of Maringá, campus Umuarama/PR that received orally the therapeutic dose of the active ingredient in question (100mg/kg). Fifty-three sheep had been treated with the same formulation/solution and no side effects were observed in any of these sheep medicated with triclorfon. But from 20 goats medicated with trichlorfon, eight goats showed, about 40 minutes after its administration, the classic clinical signs of ataxia, external lateral decubitus, drooling, tremors, constricted pupils, noisy dyspnea, involuntary urination and defecation, spastic paresis, bloat and tearing. Almost immediately after the detection of these signals, eight goats were medicated with 1% atropine sulfate (0.5mg/kg and fluid). Five of these goats received a second dose of atropine sulfate one hour after the first application because of some clinical signs such as muscle tremor still being present. Forty-eight to 72 hours after administration of trichlorfon, three of those five goats died. At necropsy we observed cyanotic mucous membranes, congestion of liver, spleen, kidneys and mesenteric vessels, filled gallbladder, emphysema, and reddish lungs. The results of this study call attention to the outbreak that occurred in adult goats in good clinical conditions and, above all, that they received the therapeutic dosage recommended on the manufacture label. This suggests a higher sensitivity of the species to the recommended dose of trichlorfon (100mg/kg) for goats.

E-mail: wdzlopes@hotmail.com O presente estudo descreve um surto de intoxicação por triclorfon em caprinos, que receberam pela via oral, a dosagem terapêutica do princípio ativo em questão (100mg/kg), provenientes da Universidade Estadual de Maringá, campus de Umuarama, PR. Cinquenta e três ovinos foram tratados com a mesma formulação/solução e não se observou nenhum efeito colateral nos animais referente ao tratamento com triclorfon. Das 20 cabras medicadas, cerca de 40 minutos após a administração do triclorfon, oito apresentaram os clássicos sinais clínicos de ataxia, decúbito externo-lateral, sialorreia, tremores, constrição das pupilas, dispneia com ruídos, micção e defecação involuntária, paresia espástica, timpanismo e lacrimejamento. Quase que imediatamente após a detecção destes sinais, as oito cabras foram medicadas com sulfato de atropina 1% 0,5mg/kg mais fluidoterapia. Cinco destes animais tiveram de receber nova dosagem de sulfato de atropina uma hora após a primeira aplicação, em função de alguns sinais clínicos, como tremores musculares, ainda estarem presentes. De 48 a 72 horas após a administração do triclorfon, três destes cinco animais vieram a óbito. Na necropsia, foi possível observar mucosas cianóticas, congestão de fígado, baço e rins, vasos mesentéricos congestos, vesícula biliar repleta, enfisema pulmonar, parênquima pulmonar avermelhado. Os resultados encontrados neste trabalho chamam atenção que o surto aconteceu em cabras adultas, que apresentavam boas condições clínicas e acima de tudo, receberam a dosagem terapêutica recomendada em bula pelo fabricante. Talvez isso possa indicar alguma sensibilidade mais elevada desta espécie animal à dosagem recomendada em bula do triclorfon (100mg/kg) para caprinos.

E-mail: asborges@fmvz.unesp.br Poisoning by monosodium methanearsonic acid (MSMA) is reported in a herd of 24 Girolando cows that were introduced into a pasture sprayed with the herbicide. Clinical signs were apathy, anorexia, and profuse diarrhea. Fourteen cows died and two were necropsied. Abomasal ulcers and renal congestion was observed. Main histologic lesions were multifocal, accentuated, necrotizing and hemorrhagic abomasitis and omasitis, and tubular necrosis in the kidneys. Mean arsenic concentrations in cows with clinical signs were 1.19±0.40, 10.52±2.16, and 76.06±48.37ppm in blood, milk, and feces, respectively. In the two necropsied cows arsenic concentrations were 25.58 and 23.85ppm in liver, and 28.71 and 35.94ppm in kidney, respectively. In a fetus of a cow that was necropsied, arsenic concentrations were 9.0 and 8.92ppm in liver and kidney, respectively. Arsenic concentration in the grass collected from the paddock sprayed with MSMA was 111.58ppm. In Brazil, the use of MSMA in the composition of herbicides is allowed only for agricultural use, not for livestock. The use of arsenic based products for livestock can lead to high mortality rates in the herd, as well as reduced production and contamination of animal products.

Os principais achados macroscópicos foram úlceras abomasais e congestão renal. No exame microscópico, as principais lesões observadas foram abomasite e omasite necro-hemorrágica multifocal acentuada e, nos rins, necrose tubular difusa. As concentrações médias de arsênico em vacas com sinais clínicos foram 1,19±0,40, 10,52±2,16 e 76,06±48,37ppm no sangue, leite e fezes, respectivamente. Os níveis de arsênico encontrados em dois animais necropsiados foram 25,58 e 23,85ppm em fígado, e 28,71 e 35,94ppm em rins, respectivamente. No feto de uma vaca necropsiada, os níveis de arsênico mensurados no fígado e rim foram 9,0 e 8,92ppm, respectivamente. A concentração de arsênico no capim do piquete pulverizado foi 111,58ppm. No Brasil, o uso MSMA na composição de pesticidas e herbicidas é permitido somente para uso agrícola, mas não pecuário. A utilização desse ou de outros produtos à base de arsênico na pecuária pode causar altos índices de mortalidade no rebanho, além de diminuição da produção e contaminação de produtos de origem animal.


#5 - Renal cortex copper concentration in acute copper poisoning in calves, 32(1):1-4

E-mail: fazzio@fcv.unlp.edu.ar The aim of this study was to estimate the diagnostic value of renal cortex copper (Cu) concentration in clinical cases of acute copper poisoning (ACP). A total of 97 calves that died due to subcutaneous copper administration were compiled in eleven farms. At least, one necropsy was conducted on each farm and samples for complementary analysis were taken. The degree of autolysis in each necropsy was evaluated. The cases appeared on extensive grazing calf breeding and intensive feedlot farms, in calves of 60 to 200 kg body weight. Mortality varied from 0.86 to 6.96 %, on the farms studied. The first succumbed calf was found on the farms between 6 and 72 hours after the susbcutaneous Cu administration. As discrepancies regarding the reference value arose, the local value (19.9 parts per million) was used, confirming the diagnosis of acute copper poisoning in 93% of the analyzed kidney samples. These results confirm the value of analysis of the cortical kidney Cu concentration for the diagnosis of acute copper poisoning.

Um total de 97 bezerros foi compilado em onze fazendas. Pelo menos, uma necropsia foi realizada em cada caso e foram colhidas amostras para análise complementar. O grau de autólise em cada necropsia foi avaliado. Os casos aparecem em criação extensiva e também em fazendas de confinamento intensivo. Os pesos dos animais variavam de 60 até 200 kg. Mortalidade variou entre 0,86 e 6,96%, em todas as fazendas estudadas, o primeiro animal morto foi observado entre 6 e 72 horas após à administração parenteral de Cu. Surgirem discrepâncias em relação ao valor de referência a ser usado. O valor local (19. 9 partes por milhão) foi usado, confirmando o diagnóstico de intoxicação aguda de cobre em 93% das amostras analisadas nos rins. Estes resultados confirmam o valor diagnóstico da concentração de Cu no rim córtex para o diagnóstico de ACP.

E-mail: moleta@ufmt.br Hepatocellular necrosis are reported in swine after consumption of diets containing grains of “sorgo-granífero” (Sorghum bicolor) accidentally contaminated with Crotalaria spectabilis seeds in the municipality of Juscimeira, MT. Clinical signs began 24-48 hours after consumption of contaminated ration and were characterized by depression, lethargy, apathy, loss of appetite, vomiting, pale or jaundiced mucous membranes, ascites, lateral recumbency, the lateral position with paddling and convulsions, clinical outcome was 48-60 hours followed by death. 76 pigs died in four properties. The main gross lesions were liver increased in size and lobular illustration with red-brown central areas interspersed with yellowish areas, ascites and hydrothorax with reddish-yellow liquid containing filaments with aspect of fibrin, enlarged lymph nodes and interlobular pulmonary edema. The disease was experimentally reproduced with 16 pigs divided into six groups that received seeds of C. spectabilis in different doses. Hepatocellular necrosis occurred in seven pigs, two of which received daily doses of 2.5g/kg and five who received single doses of 5 and 9.5g/kg. Ten daily doses of 0.5 and 1.25g/kg caused liver fibrosis.

As Principais alterações macroscópicas foram fígado aumentado de tamanho com evidenciação do padrão lobular, ascite e hidrotórax com líquido de coloração amarelo avermelhado contendo filamentos com aspecto de fibrina, linfonodos aumentados e edema pulmonar interlobular. A doença foi reproduzida utilizando-se 16 suínos divididos em seis grupos que receberam sementes de C. spectabilis em diferentes doses. Necrose hepatocelular ocorreu em sete suínos, sendo dois que receberam doses diárias 2,5g/kg e cinco que receberam doses únicas de 5,0 e 9,5g/kg. Dez doses diárias de 0,5 e 1,25g/kg causaram fibrose hepática.

Mimosoideae).] Intoxicação aguda e abortos em cobaias pelas favas de Enterolobium contortisiliquum (Leg. Mimosoideae). Pesquisa Veterinária Brasileira 28(12):593-596. E-mail: bonel-raposo@brturbo.com.br The objective was to study the acute toxicity and the abortive properties of Enterolobium contortisiliquum pods in guinea pigs. Pods of E. contortisiliquum were administered orally to 4 groups of 3 guinea pigs each. Another group of 3 guinea-pigs was used as control. Group 1 and 2 were fed with one dose of 5 and 10g of pods for kg body weight, respectively. The guinea pigs of Group 3 and 4 received 10 and 15g/kg, respectively, divided into daily doses of 5g/kg. One guinea pig from Group 2 and one from Group 4 died 12 and 18 hours after the end of the administration. Gross lesions were hemorrhages of the stomach and of the large and small gut, enlarged liver, and dilated gall bladder. Histologically, the liver had severe vacuolation and necrosis of periportal hepatocytes. In another experiment a ration containing 4% of pods of E. contortisiliquum was fed to 2 groups of 4 guinea-pigs, 35 days after mating. Four of the 8 guinea pigs aborted 6-15 days after the beginning of ingestion. The other 4 guinea pigs were not pregnant. All guinea pigs were euthanized after abortion or at the end of the experiment. Histologically all animals had mild to severe periportal hemorrhagic necrosis. All fetuses had variable degree of autolysis. In 4 fetuses studied no significant histologic lesions were observed. The acute lesions observed in guinea-pigs are similar than those observed in the spontaneous poisoning by Enterolobium spp. in cattle. Similar lesions are observed in guinea-pigs poisoned experimentally with saponins from E. gummiferum. The results of the experiments in pregnant guinea pigs suggest that E. contortisiliquum can be used to study the abortive effect of its pods or its toxic compounds.

E-mail: bonel-raposo@brturbo.com.br The objective was to study the acute toxicity and the abortive properties of Enterolobium contortisiliquum pods in guinea pigs. Pods of E. contortisiliquum were administered orally to 4 groups of 3 guinea pigs each. Another group of 3 guinea-pigs was used as control. Group 1 and 2 were fed with one dose of 5 and 10g of pods for kg body weight, respectively. The guinea pigs of Group 3 and 4 received 10 and 15g/kg, respectively, divided into daily doses of 5g/kg. One guinea pig from Group 2 and one from Group 4 died 12 and 18 hours after the end of the administration. Gross lesions were hemorrhages of the stomach and of the large and small gut, enlarged liver, and dilated gall bladder. Histologically, the liver had severe vacuolation and necrosis of periportal hepatocytes. In another experiment a ration containing 4% of pods of E. contortisiliquum was fed to 2 groups of 4 guinea-pigs, 35 days after mating. Four of the 8 guinea pigs aborted 6-15 days after the beginning of ingestion. The other 4 guinea pigs were not pregnant. All guinea pigs were euthanized after abortion or at the end of the experiment. Histologically all animals had mild to severe periportal hemorrhagic necrosis. All fetuses had variable degree of autolysis. In 4 fetuses studied no significant histologic lesions were observed. The study encompassed 6,256 necropsies of cattle considering a 43-year period and included 15 necropsies of cattle dying from acute bracken poisoning. Morbidity and mortality were 17.9% and lethality was virtually 100%. In 40% of the farms the disease occurred in small outbreaks affecting several cattle per farm and in 60% only one bovine was affected in each farm. Main clinical signs include fever (40-42°C), apathy, drooling, and hemorrhages; the latter were mainly from the gums, nostrils, and gastrointestinal tract. Multiple petechiae were observed in several mucosae and in the skin. Occasionally hematuria and blood in the milk were reported. The disease was invariably fatal after a course of approximately two days. Hematological changes included severe neutropenia, non-regenerative normocytic normochromic anemia and thrombocytopenia. Necropsy findings include hemorrhages o varying degrees in several organs and infarcts in the liver. Both hemorrhages and hepatic infarcts were confirmed histologically; clusters of bacterial rods and thrombosed vessels were associated with the infarcts. Marked bone marrow aplasia was a consistent finding in the four cases in which the marrow was evaluated.

Clinical manifestations of the natural poisoning include muscle tremors, hypersalivation, groaning and rapid death. In the experimental poisoning lateral recumbency, dyspnea, tetanic spasms and paddling was also observed. The main necropsy findings were reddening, edema and ulceration of the ruminal and abomasal mucosa. Histopathological findings were necrosis of the epithelium of the forestomachs with polymorphonuclear infiltration of the submucosal and muscular layers. Tubular renal necrosis, liver portal necrosis and necrosis of the lymphoid tissues was also noted. The sodium fluorsilicate caused clinical signs at a dose of 300mg/kg, and death from 400mg/kg on.
